Ketorolac Tromethamine is used for the short-term treatment of moderate to severe pain in adults. It is usually used before or after medical procedures or after surgery. Reducing pain helps you recover more comfortably so that you can return to your normal daily activities. This medication is a nonsteroidal anti-inflammatory drug (NSAID). It works by blocking your body’s production of certain natural substances that cause inflammation. This effect helps to decrease swelling, pain, or fever.Ketorolac Tromethamineshould not be used for mild or long-term painful conditions (such as arthritis).

Use the lowest effective dose for the shortest duration consistent with individual patient treatment goals.Ketorolac Tromethamine is a Non-steroidal anti-inflammatory drug (NSAID) and has antipyretic, analgesic and anti-inflammatory properties. It is indicated for short term management of acute pain that requires the calibre of pain management offered by opioids. Clinicians may choose to initiate ketorolac tromethamine to manage post-operative pain, spinal and soft tissue pain, rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, menstrual disorders and headaches among other ailments. Regardless of the etiology of pain, patients should use the lowest possible dose, and avoid using ketorolac tromethamine for an extended period of time (ideally ≤ 5 days). A benefit of choosing ketorolac tromethamine over other analgesics with similar potency is that that there does not appear to be a risk of dependence or tolerance with ketorolac tromethamine use.
Ketorolac tromethamine is contraindicated in patients with active peptic ulcer disease, in patients with recent gastrointestinal bleeding or perforation and in patients with a history of peptic ulcer disease or gastrointestinal bleeding.

Ketorolac tromethamine is a nonsteroidal anti-inflammatory drug (NSAID) that exhibits analgesic activity in animal models. The biological activity of ketorolac tromethamine is associated with the S-form. Ketorolac tromethamine possesses no sedative or anxiolytic properties. The peak analgesic effect of Toradol occurs within 2 to 3 hours and is not statistically significantly different over the recommended dosage range of Toradol. The greatest difference between large and small doses of Toradol is in the duration of analgesia.
Ketorolac tromethamine is a racemic mixture of [-]S- and [+]R-enantiomericforms, with the S-form having analgesic activity. In adults, the extent of bioavailability following administration of the oral form of Toradol and the IM form of ketorolac tromethamine was equal to that following an IV bolus.
Mechanism of action :
Ketorolac tromethamine inhibits key pathways in prostaglandin synthesis which is crucial to it’s mechanism of action. Although ketorolac tromethamine is non-selective and inhibits both COX-1 and COX-2 enzymes, it’s clinical efficacy is derived from it’s COX-2 inhibition. The COX-2 enzyme is inducible and is responsible for converting arachidonic acid to prostaglandins that mediate inflammation and pain. By blocking this pathway, ketorolac tromethamine achieves analgesia and reduces inflammation. Ketorolac tromethamine is administered as a racemic mixture; however, the “S” enantiomer is largely responsible for it’s pharmacological activity.
